This works on my system, which is Slackware 10.2.

bash-3.00$ cat /etc/slackware-version 
Slackware 10.2.0
bash-3.00$ grep RTF_UP /usr/include/net/route.h
#define RTF_UP          0x0001          /* Route usable.  */

Are you missing /usr/include/net/route.h?  It's in glibc-2.3.5-i486-5 for me.

bash-3.00$ grep route.h /var/log/packages/*
/var/log/packages/glibc-2.3.5-i486-5:usr/include/net/route.h
