Like most (all?) games in the RE series, RE5 needs wmp10 installable by winetricks, followed by windowsmedia-kb942423-x86-intl.exe to run. After that, the game launches fine. Installing d3dcompiler_43 d3dx9 and xact_jun2010 as per the appdb page is no longer necessary. There's been quite a lot of progress over the last couple of years.
