Since several Wine versions many - not say nearly all but one - of mine Windows applications stopped working. I have now tested and found that V1.5.24 works perfectly e.g. with Dragon Age II, while 1.6-rc4 doesn't even let me to the configuration screen. Same prefix. Same compilation options (I use Gentoo, therefore I can change, and I tried, but without any success...)
