So basically, all games running RPG maker VX or VX ACE games by using the RTP will crash. This has been an issue since 1.9 and continues to exist in 2.1. Install the RTP itself will run fine without any error messages. The crashes occur after selecting "new game" in the startup menu of any RPG maker game. Trying to load a save file does not fix the issue. These crashes do not occur in 1.8.6 which is the version that I have been forced to use because of these issues. Is there any sample/demo/freeware game that can reproduce the issue?
The crash is inside winegstreamer so you can try disabling it in winecfg. Open winecfg -> libraries. Then add winegstreamer and edit and select disable.

I can't confirm this bug in wine-2.1, 3.0-rc2, 3.0-rc5 and staging 2.20. I used the game Crysalis from the link in Comment #2. It uses VX ACE RTP. Yes, I didn't change anything. Everything gstreamer related is installed and while running the intro movie, fixme's about gstreamer stubs showed up (not errors about missing plugins). Even in the latest version of WINE I can reproduce this bug. Disabling WINEGStreamer fixes the issue completely. So I don't consider it a major issue. More of a slight annoyance. It should also be noted that RPG Maker VX doesn't work in WINE and will cause a black screen issue. The Audio will actually work completely fine. You can even select new game while the screen is black. But the graphics fail to render. The RPG Maker VX error is easily fixed by simply enabling a desktop. I found the same error on another game called "Siberia." And the person solved it by using a desktop. I installed the VX ACE RTP from http://www.rpgmakerweb.com/download/additional/run-time-packages, installed Crysalis from http://www.rpgmakerweb.com/download/additional/sample-games, and ran the game. I selected "New Game" and the game played its audio successfully without crashing. This is with 32-bit gstreamer1.0 version 1.8.3. Can you please try a fresh wineprefix, and, if you are still experiencing this bug, post a log with +quartz,+gstreamer?
