This may result in unexpected behaviours. Do you want to launch the game anyway?" Game launches normally afterwards. Strange, don't have this with 6600GT. Confirming per comment 2.
Maybe this only occurs for series 7 thru 9 cards? Without that (with default being backbuffer) I'm getting the same message. I'm not sure there is anything could be done here. the R5G6B5 mode is a 16-bit mode. Wine can't switch BPP since X does not support that. And backbuffer works with the screen color depth only.
