First, I'm using wine-staging 1.9.18 from the wine-builds ppa on Lubuntu 16.04 LTS.
Second, Like the reporter, I discovered this in a 64-bit wineprefix. I then retried on a 32-bit wineprefix, but the error persists. Both prefixes are clean, except for vcrun2010 (installed by winetricks), which is required by Desmume.
Finally, unless you want to see what Desmume itself has to say on startup, console output won't tell you any more than the attached error report does, at least with the default WINEDEBUG settings, because the only console output from this is what Desmume itself outputs, and the error report itself (illegal access exception).
If you need further debugging output, I'll need to know what to put in WINEDEBUG for you, so that you get proper console output.
One last thing: Given the winehq history on this, it's very possible that this is a regression and/or a problem with ubuntu. Unfortunately, I'm not in a position to test either theory, as I don't have a build environment for wine set up atm.

P.S. Desmume's linux build works alright, but the GUIs are seriously behind the windows version.
