libjpeg62 is available on (X)Ubuntu LTS 20.04 and I installed it. libjpeg62:i386 is not available.

Pure 64-bit Wine can only run pure 64-bit windows applications, which are very uncommon and currently unsupported. https://forum.winehq.org/viewtopic.php?t=33427&p=126290
Shared wow64 Wine build (the default on 64-bit systems) still requires 32-bit dependencies to run 32-bit binaries.
libjpeg62:i386 is available on Ubuntu 20.04: https://packages.ubuntu.com/focal/libjpeg62
Synaptic won't display i386 packages in the default view, if that's what you tried. If you need help installing i386 architecture packages on ubuntu, please check ubuntu forums: https://ubuntuforums.org/showthread.php?t=2418075&p=13856444#post1385644...
Since it works with vanilla wine, it is either an issue of missing dependency or a playonlinux issue. I'll mark the bug as invalid. Feel free to reopen if it can be reproduced with vanilla Wine.
