After doing a new install of desura in a clean prefix, I get the following
error trying to start it: "Failed to update Desura: Check sum failed on file
\desura.exe [49.0]"

After running it for the first time desura.exe is replaced by an empty file, so
I can't try to start it at all.

Are you sure that it wasn't simply a download fail (maybe an unstable internet
connection or a server error)?
I tried it on my own with newest wine and Desura just works.
Could you please reinstall and retest it with current wine?

I tried the exe with 1.7.22

and it updates for me.

I think too that this is invalid.

Possible ... if you have too much time, test with Wine 1.5.25

$ sha1sum DesuraInstaller.exe 
8694863a4fe8989de4e57d249ba23e637dc36cd7  DesuraInstaller.exe

$ du -sh DesuraInstaller.exe 
1.2M    DesuraInstaller.exe

$ wine --version
wine-1.7.22-70-g509bdae
