actually my system is:
kernel version: 2.6.18-5-k7 #1 SMP
wine: wine-0.9.49
nvidia driver version: 2.6.18-5-k7 SMP mod_unload K7 REGPARM gcc-4.1
i have the same problem:
wine: wineprefixcreate failed while creating '/root/.wine'.
