Program Error popup shows up as soon as Mapsource is launched. Mapsource starts and can be used.

There is something wrong with your wine prefix, based on the log builtin mscoree is used although you have installed .NET 4.0. Please set mscoree to "native" in the winecfg libraries tab.
If that doesn't help you should start again with a fresh wineprefix and use winetricks for the .NET installation ("winetricks dotnet40").

Thanks for the quick reply and your analysis. Indead, setting mscoree to native solved the problem. So, my bad as I went through a remove/reinstall Wine procedure. Problem solved.

Glad that it works now. :) Nothing was fixed in Wine, so I'm changing the status to INVALID.
