When running the Myst V installer it comes up with a message saying that it cannot run in graphical mode and exits.

The file you are trying to attach is 19478 kilobytes (KB) in size. Non-patch attachments cannot be more than 1000 KB. We recommend that you store your attachment elsewhere and then specify the URL to this file on the attachment creation page in the AttachURL field. Alternately, if your attachment is an image, you could convert it to a compressible format like JPG or PNG and try again.

Eh? The URL you have points to a 3.4KB file. Also, creating an attachment that contains just a text link is just silly. Put the link as a comment if you absolutely must.
Furthermore, how are you generating such a large file? Are you passing any flags to wine? If so, don't.

Don't do that unless you're asked to. Just run wine setupwin32.exe >& ~/output.txt and attach output.txt here. If it's too large, bzip2 it: $ bzip2 ~/output.txt and attach ~/output.txt.bz2

The demo seems to have a different problem it gets to the same point as the full game, but instead of saying it can't run in graphical mode it just draws an empty dialogue box.

It still is a problem with wine 1.6rc3. I tried this with a freshly created wineprefix (WINEARCH=win32 - I'm on a x86_64 system). As a second step I tried with native msvcrt.dll, msvcp71.dll and msvcr71.dll - got them with "winetricks vcrun6 vcrun2003". In both cases the installation seems to start fine, but then aborts with a dialog box saying that the installation program cannot run in GUI mode. Output for both tests is attached.
Installation seems to work when running "wine setupwin32.exe -silent". The game still does not work, but that might not be related...
